I want to know is it possible to takeoff in PositionHold mode.??
Yes, this requires a few things to be working before that will work. Compass needs a good position fix or it will refuse to arm. The flight controller needs to be flashing green.

Yup. You need 3D GPS lock, calibrated compass and the usual checks.
