Question about Autotune

Hello, I have a few questions about the PID setting.

1. If I make the bank angle 50 degrees, do I need to autotune again?
2. During the flight, there will be a weight change of 300 gr. According to the information I received from the forum, I did the autotune when there was a low weight, does it cause a problem?
